dc.description.abstractA presente tese foi realizada com o objetivo de avaliar três diferentes ambientes naturais, utilizando ovos e filhotes recém eclodidos das espécies de testudines, Podocnemis expansa, Chelonia mydas e Caretta caretta, como bioindicadores da presença de metais em ecossistemas marinho e de água doce. Para tal foram realizadas quantificações de metais em ovos coletados no momento da postura (sem contato com o sedimento) e em filhotes recém eclodidos após cerca de 60 dias de incubação, assim como em amostras do sedimento dos ninhos. Dessa forma foi possível averiguar a transferência de metais de origem materna e transferência a partir do sedimento após incubação. Baseada nessas análises a tese foi estruturada em três diferentes capítulos. O primeiro capítulo relata a quantificação de metais (Fe, Mn, Ni, Pb e Cd) em ovos e filhotes recém eclodidos da tartaruga C. mydas nas praias da Ilha de Trindade, onde foi identificada a transferência materna de metais, assim como foi observado um aumento na quantidade desses metais provenientes do sedimento. Embora tenha havido aumento na quantidade de metais nas amostras após incubação, os valores ainda se apresentam baixos, sem haver evidências de causarem danos genotóxicos aos filhotes, visto que a frequência de micronúcleo (MN) eritrocitária foi baixa (1,25 ‰). Esses resultados indicam um ambiente insular ainda conservado. No segundo capítulo, foi realizada a avaliação toxicológica comparativa da bioacumulação de metais (Al, Cd, Co, Cr, Cu, Fe, Mn, Pb e Zn) em ovos e filhotes recém eclodidos de tartarugas de água doce, P. expansa, incubadas nas areias das praias de dois rios: Araguaia (ambiente protegido da Área de Proteção Ambiental Meandros do Araguaia) e Crixás-Açu, que é menor e que deságua no rio Araguaia. Nesse capítulo, pode-se observar que há transferência materna de metais e que os dois ambientes estão afetados por metais, visto que houve um acúmulo significativo de metais nos ovos e filhotes de P. expansa. Contudo, a região do rio Crixás-Açu encontra-se mais afetada, uma vez que as amostras coletadas nesse local apresentam concentrações maiores de metais, e consequentemente uma frequência de MN maior (15 ‰), em contraposição aos valores de 10 ‰ de MN nos filhotes incubados nas areias do rio Araguaia. No terceiro capítulo, foi feito o biomonitoramento de três anos consecutivos da área da foz do Rio Doce após ter sido atingida pela lama de rejeitos proveniente do rompimento da barragem da mineradora Samarco em 2015. Nesse capítulo foram utilizados ovos e filhotes recém eclodidos da espécie de tartaruga marinha C. caretta. Foram realizadas coletas na temporada reprodutiva da espécie de 2015/2016; 2016/2017 e 2017/2018, houve ainda uma coleta adicional de sedimento em novembro de 2015 antes da lama atingir a foz do rio. Os resultados indicam que os metais mensurados (Ag, As, Cd, Co, Cr, Cu, Fe, Hg, Mn, Ni, Sb, Sn, Zn) já se encontravam presentes no sedimento da região antes mesmo do desastre acontecer, sendo acumulados nos ovos e filhotes; assim como houve transferência materna de metais. Contudo esses metais causaram danos genotóxicos maiores nos filhotes coletados na temporada de 2015/2016 (4,52 ‰ de MN); assim como um maior acúmulo de alguns metais nessa primeira coleta. No decorrer dos anos de biomonitoramento, o ambiente de incubação das praias se mostrou menos tóxico para a incubação desses filhotes do ponto de vista dos danos genotóxicos, em virtude de uma diminuição significativa na frequência de micronúcleos eritrocitários (2,84 ‰ em 2016/2017 e 1,47 ‰ em 2017/2018). Em nenhum dos anos 3 anos de biomonitoramento foram observadas no tecido hepático dos filhotes alterações na atividade enzimática da glutationa S-transferase e da Catalase. Como conclusão geral, este estudo identificou a presença de metais em cascas de ovos e filhotes das três espécies de tartarugas sejam de água doce ou marinha, bem como nos sedimentos das três áreas de biomonitoramento. Os metais encontrados nas cascas de ovos e nas gemas de ovos resultaram da transferência materna, enquanto o aumento dos metais que ocorrem nas cascas de ovos após a incubação e os tecidos dos filhotes foram transferidos dos sedimentos para os ninhos. Contudo obtivemos um ambiente insular que podemos considerá-lo controle para as outras áreas que são impactadas por ações antrópicas.pt_BR

dc.description.resumoThe present thesis was carried out with the objective of evaluating three different natural environments, using eggs and newly hatched chicks of the species of testudines, Podocnemis expansa, Chelonia mydas and Caretta caretta, as bioindicators of the presence of metals in marine and freshwater ecosystems. For this purpose, quantifications of metals were performed in eggs collected at the time of laying (without contact with the sediment) and in newly hatched chicks after about 60 days of incubation, as well as in samples of the nest sediment. In this way it was possible to verify the transfer of metals of maternal origin and transfer from the sediment after incubation. Based on these analyzes, the thesis was structured in three different chapters. The first chapter reports the quantification of metals (Fe, Mn, Ni, Pb and Cd) in eggs and young hatchlings of the turtle C. mydas on the beaches of Trindade Island, where maternal metal transfer was identified, as was observed an increase in the amount of these metals from the sediment. Although there was an increase in the amount of metals in the samples after incubation, the values are still low, with no evidence of causing genotoxic damage to the young, since the frequency of erythrocyte micronucleus (MN) was low (1.25 ‰). These results indicate an island environment still conserved. In the second chapter, a comparative toxicological evaluation of the bioaccumulation of metals (Al, Cd, Co, Cr, Cu, Fe, Mn, Pb and Zn) was carried out on eggs and hatchlings of freshwater turtles, P. expansa, incubated on the sands of the beaches of two rivers: Araguaia (protected environment of the Meandros do Araguaia Environmental Protection Area) and Crixás-Açu, which is smaller and flows into the Araguaia River. In this chapter, it can be observed that there is a maternal transfer of metals and that both environments are affected by metals, since there was a significant accumulation of metals in the eggs and chicks of P. expansa. However, the region of the Crixás- Açu river is more affected, since the samples collected in this place have higher concentrations of metals, and consequently a higher frequency of MN (15 ‰), in contrast to the values of 10 ‰ of MN in the hatchlings incubated in the sands of the Araguaia river. In the third chapter, biomonitoring was carried out for three consecutive years in the area of the mouth of the Rio Doce after being hit by the tailings mud from the rupture of the Samarco mining dam in 2015. In this chapter, newly hatched eggs and hatchlings of the species of sea turtle C. caretta. Collections were carried out in the 2015/2016 species breeding season; 2016/2017 and 2017/2018, there was an additional sediment collection in November 2015 before the mud reached the mouth of the river. The results indicate that the metals measured (Ag, As, Cd, Co, Cr, Cu, Fe, Hg, Mn, Ni, Sb, Sn, Zn) were already present in the region's sediment even before the disaster happened, being accumulated in eggs and chicks; just as there was a maternal transfer of metals. However, these metals caused greater genotoxic damage in the pups pollected in the 2015/2016 season (4.52 ‰ of MN); as well as a greater accumulation of some metals in this first collection. During the years of biomonitoring, the beach incubation environment proved to be less toxic for the incubation of these young from the point of view of genotoxic damage, due to a significant decrease in the frequency of erythrocyte micronuclei (2.84 ‰ in 2016/2017 and 1.47 ‰ in 2017/2018). In none of the 3 years of biomonitoring were changes in the liver tissue of the puppies observed in the enzymatic activity of glutathione S-transferase and Catalase. As a general conclusion, this study identified the presence of metals in egg shells and hatchlings of the three species of turtles, whether freshwater or marine, as well as in the sediments of the three biomonitoring areas. The metals found in egg shells and egg yolks resulted from maternal transfer, while the increase in metals that occur in egg shells after incubation and the tissues of the chicks were transferred from the sediment to the nests. However, we obtained an island environment that we can consider as control for the other areas that are impacted by anthropic actions.pt_BR
