dc.description.abstractAs algas marinhas são ricas em ácidos graxos e polifenóis que possuem propriedades antioxidantes e anti-inflamatórias, promovendo o reparo de lesões. Poucas são as alternativas para o tratamento de lesões cutâneas, especialmente no que diz respeito à melhora no aspecto global da lesão. Sendo assim, o presente estudo teve como objetivo avaliar o efeito da pomada de Padina na cicatrização de feridas cutâneas in vivo. Ao todo, 52 animais tiveram o dorso tricotomizado e quatro feridas foram feitas na pele de cada animal usando um punch de 15 mm. Na sequência, os animais foram tratados topicamente com pomada de Padina a 25% (Grupo P; n = 20); controle Lanovaselina com sal (Grupo LS; n = 20) ou controle Lanovaselina sem sal (Grupo L; n = 12). Todos os animais foram tratados diariamente logo após a lesão. Após 3, 7, 14 e 21 dias da cirurgia, os animais foram eutanasiados com sobredose de anestésico (n = 5/grupo) e as feridas foram fotografadas para a determinação das taxas de fechamento. As feridas foram excisadas e armazenadas a -80ºC ou em tampão formalina para as análises bioquímicas e histológicas, respectivamente. Para a análise histológica, as lâminas foram desidratadas, fixadas e coradas com hematoxilina/eosina (H/E) ou Picrosirius red e analisadas em microscópio óptico. As lâminas foram fotomicrografadas e posteriormente analisadas utilizando o software Image J® para determinação da deposição de colágeno e análise do infiltrado inflamatório. Os tecidos congelados foram homogenizados e a atividade da Mieloperoxidase (MPO) e da N-acetil-β-D-glicosaminidase (NAG) foram determinadas utilizando técnica colorimétrica. O tratamento com Padina evidenciou maior retração da ferida nos dias 3 e 7 quando comparado ao grupo lanovaselina sem sal. Na quantificação do colágeno foi observada a redução na deposição do colágeno após 7 dias de tratamento com Padina. O resultado do infiltrado inflamatório revelou que o tratamento com Padina foi capaz de reduzir o infiltrado inflamatório nos dias 3, 7 e 14 da indução da lesão. Esses resultados foram acompanhados de redução na atividade de MPO após 7 dias e de NAG após 7 e 14 dias do tratamento com Padina quando comparado ao grupo lanovaselina sem sal. Deste modo, podemos concluir que o tratamento com Padina acelerou o processo de cicatrização in vivo, e que essa atividade pode ser atribuída, pelo menos em parte, à redução na resposta inflamatória tecidual.pt_BR

dc.description.resumoSeaweed is rich in fatty acids and polyphenols that have antioxidant and anti-inflammatory properties, promoting injury repair. There are but few alternatives for the treatment of skin lesions, especially regarding the improvement of the global aspect of the lesion. Thus, the aim of this study was to evaluate the effect of Padina ointment on wound healing in vivo. In all, 52 animals were trichotomized on the back and four wounds were made on the skin of each animal using a 15 mm punch. Following, the animals were treated topically with P. gymnospora ointment at 25% (Group P; n=20); Salted lanovaselin (Group LS; n=20) or Unsalted lanovaselin (Group L; n=12). All animals were treated daily. At 3, 7, 14 and 21 days after surgery, the animals were euthanized with an overdose of anesthetic (n=5/group) and the wounds were photographed to determine closure rates. The wounds were excised and stored at -80 °C or in a formalin buffer for histological and biochemical analysis, respectively. For the histological analysis, slides were dehydrated, fixed and stained with hematoxylin / eosin (H / E) or Picrosirius red and analyzed under an optical microscope. The slides were photomicrographed and subsequently analyzed using Image J® software for the determination of collagen deposition and for the analysis of the inflammatory infiltrate. Frozen tissues were homogenized and myeloperoxidase (MPO) and N-acetyl-β-D-glycosaminidase (NAG) activity were determined using colorimetric technique. The treatment with Padina evidenced greater wound retraction on days 3 and 7 when compared to the Unsalted lanovaselin group. Collagen quantification showed a reduction in collagen deposition after 7 days of treatment with Padina. The result of the inflammatory infiltrate revealed that the treatment was able to reduce the inflammatory infiltrate on days 3, 7 and 14 of lesion induction. These results were accompanied by a reduction in MPO activity after 7 days of treatment, and of NAG after 7 and 14 days of treatment with Padina when compared to the Unsalted lanovaselin group. Thus, we can conclude that the treatment with Padina accelerated the healing process in vivo, and that this activity may be attributed, at least in part, to the reduction in the inflammatory tissue response.pt_BR
